Abstract

Study aims to determine the hydrolysis reaction of paracetamol at 37 oC in a
solution of pH 2.0; 5.0; and 7.0 and to determine the levels of p-aminophenol
contained in the raw material of paracetamol. This research was conducted
Qualitative and Quantitative using the HPLC method, the stationary phase in
the form of column L7 (4.6 mm x 25 cm) and the mobile phase in the form of
solution A - solution B (20: 80). The p-aminophenol standard was dissolved
using the mobile phase at a concentration of 0.1 mg/mL. Test solutions with
buffer pH 2.0; 5.0; and 7.0 were first incubated at 37 oC. Determination of paminophenol levels by HPLC at a wavelength of 300 nm with a pump pressure
of 1 mL/minute with an injection volume of 5 µL. The results of Qualitative test
on samples of paracetamol raw materials showed a hydrolysis reaction of
paracetamol in a solution of pH 2.0; 5.0; and 7.0. While the results of the
Quantitative test on the raw material sample of paracetamol with a concentration
per 100 mg/mL triplo injection at pH 2.0; 5.0; and 7.0, the result is pH 2.0, which
is 0.275%; pH 5.0 is 0.228%, and pH 7.0 is 0.045%. From these results it can
be concluded that the paracetamol sample at pH 2.0 and pH 5.0 did not meet the
requirements, while the paracetamol sample at pH 7.0 met the requirements,
namely not more than 0.05%.
